چکیده

The requirement for plastoquinone in the photosynthetic phosphorylation activity of spinach chloroplasts has been indicated in an earlier publication (1). Removal of endogenous plastoquinone from lyophilized chloroplasts by heptane extraction caused a loss of photophosphorylation activity. The photophosphorylation activity could be partially restored by addition of pure plastoquinone to the extracted preparations. The Hill reaction activity with 2,3’, 6trichlorophenol indophenol responded in a fashion similar to photophosphorylation with these preparations. It was shown that this restoration of activities could be made proportional to the concentration of plastoquinone available to the heptane-extracted chloroplasts. This paper reports in detail the techniques used in such experiments and the specificity of plastoquinone as a cofactor for photophosphorylation.

Ferredoxin Catalyzed Cyclic Photophosphorylation: Reversal of Dibromothymoquinone Inhibition by N,N,N',N'-Tetram ethyl- /j-phenylenediamine

Conditions for washed, spinach thylakoid membranes to catalyze cyclic phosphorylation using ferredoxin as the cofactor for electron transfer have been re-examined. It was found necessary to ‘'redox poise” the system; however, the best method to accomplish the poising seemed to be the use of a reductant (glucose-6-phosphate) and not by optimizing the rate of phosphorylation with 3-(3,4-dichlorop...

Plastoquinone in Photosynthesis

Dibromothymoquinone and its hydroquinone are inhibitors of non cyclic electron flow from water to NADP, anthraquinone or methylviologen. The inhibition is competetively reversed by plastoquinone. It appears that dibromothymoquinone is an antagonist of plastoquinone and that it prevents the enzymic (by the next endogenous carrier of the chloroplast electron transport chain) but not the chemical ...

Site-specific Inhibition of Photophosphorylation in Isolated Spinach Chloroplasts by Mercuric Chloride.

Photophosphorylation associated with noncyclic electron transport in isolated spinach (Spinacia oleracea) chloroplasts is inhibited to approximately 50% by low concentrations of HgCl(2) (less than 1 mumole Hg(2+)/mg chlorophyll) when the electron transport pathway includes both sites of energy coupling. Reactions involving only a part of the electron transport system can give a functional isola...

Reactions of Antisera to Lutein and Plastoquinone with Chloroplast Preparations and their Effects on Photosynthetic Electron Transport

An antiserum to lutein inhibits photosynthetic electron transport between water and potas sium ferricyanide in chloroplasts from green Nicotiana tabacum var. John William’s Breadleaf. However, electron transport between diphenylcarbazide and potassium ferricyanide is not impaired. From this it is concluded that the photochemically active carotenoid should feed its electrons into the photosynth...

Site of bicarbonate effect in Hill reaction. Evidence from the use of artificial electron acceptors and donors.

Using artificial electron donors and acceptors, it is shown here that the major HCO3- effect in the Hill reaction is after the "primary" electron acceptor (Q) of Photosystem II and before the site of action of 2,5-dibromo-3-methyl-6-isopropyl-p-benzoquinone (at the plastoquinone pool). Chloroplasts in the presence of both 3-(3',4'-dichlorophenyl)-1,1-dimethylurea, which blocks electron flow fro...
